The London 2012 gold medallist sunk the Liverpool fighter to the canvas in the fourth and he could not beat the count.
Campbell, 29, has now won 15 of his 16 professional bouts, but this was one of the more stunning victories as he continues to rise up the lightweight rankings.
Mathews is a former world title challenger and a heavy hitter.
But it was the Hull man who delivered the bigger shots in this entertaining bout.
Mathews, 33, was expected to retire after he lost to WBO lightweight champion Terry Flanagan in his 50th fight earlier this year.
